Single-cell proteomics by mass-spectrometry Cellular heterogeneity is important to biological processes, including cancer and development. However, proteome heterogeneity is largely unexplored because of the limitations of existing methods for quantifying protein levels in single cells. Effective extension of mass spectrometry-based proteomics to single cells remains challenging. Herein we combined microfluidic nanodroplet technology with tandem mass tag (TMT) isobaric labeling to significantly improve analysis throughput and proteome coverage for single mammalian cells. Recent advances in single-cell analysis have unraveled substantial heterogeneity among seemingly identical cells at genomic and transcriptomic levels.

Single-cell proteomics center We develop mass-spectrometry based technologies for quantifying thousands of proteins and their interactions across thousands of single cells. We apply these technologies to understand emerging behaviors in biological systems and to identify mechanisms that underpin health and disease. Identify adaptive phosphoproteomic signaling networks from rare subsets of single tumor and other cell types, leveraging single cell proteomics to treat the entire set of signaling pathways concurrently, and eliminate resistance and metastases.
The single cell proteomics meeting will bring together many of the pioneers in the field for sharing ideas, results, and informal discussions of future directions.

Many pressing medical challenges, such as diagnosing disease, enhancing directed stem-cell differentiation, and classifying cancers, have long been hindered by limitations in our ability to quantify proteins in single cells. Mass spectrometry (MS) is poised to transcend these limitations by developing powerful methods to routinely quantify thousands of proteins and proteoforms across many
First, single cell proteomics is not only the instrument sensitive issue. How to handle subnanogram amounts of complex protein sample to digest them and transfer them to MS is more challenging. Single cell proteomics is a rapidly evolving field. • Mass spectrometry-based proteomics identifies thousands of proteins in single cells.
Recent advances in single-cell analysis have unraveled substantial heterogeneity among seemingly identical cells at genomic and transcriptomic levels. These discoveries have urged scientists to develop new tools that are capable of investigating single cells from a broader set of “omics”.
